Anyone have experience with this tent, which is Genuine LR? Part# VPLEP0432. looks like it would work well with the REI air mattress in the back of the 110.

Looks like all the hassle of pitching a tent without any of the benefits of actually having a tent.
I've not used it but do camp a good bit and have purchased my fair share of products that were complete pants. This is ticking all the useless boxes to my eye.
I realized after posting that this is not really a tent as you say. I do not believe there is any sort of closure across the opening.
I am looking for a SUV/truck tent that will work with the rear door in the open position. Many of those I have looked at use a raised hatch to support the tent assembly and provide headroom. .

You may want to look at Kelty Waypoint Tarp, Been looking for something that is large enough to use as sun & rain shelter, and this may fit the need. There's a few more pictures on REI, but this one shows a jeep with back door open.
On reflection what I really want is something to complement car camping, i.e. having an air mattress is the back of the 110 and then being able to step out onto a tent floor with the cargo door open. The Napier Sportz SUV tent seems like the closest thing. My concern is these tents use a top hinged rear hatch or glass panel to support part of the tent. How would it work with a large door door that opens to one side?
I don't think it would make much difference with a Defender side swing door. Remember that Wrangler pictured above has both, the top glass goes up and the lower door and tire swing right just like your Defender. That is a pretty slick little setup for camping out of your car. You sort of get a little 'living room' and the back of the car is the 'bedroom' as it were.

Just so.
They make a bigger version for not much extra cost that comes with a front porch!
The rear door of the Defender is challenging, especially since you cannot open it from the inside. Have you looked into an awning with annex on one side instead?
just an example here: ARB 813208A Awning Room Accesory (Deluxe w/Floor 2000mm x 2500mm Heavy Duty) for ARB Awning 814409 2000x25000 https://a.co/d/2Hienoa
