This is an ignition system misfire code. Have you replaced plugs lately, could be bad wires. Try new plugs and wires (Magnacore) first then clear the code and see if it returns.
Is this a DI or DII?

DII, DirtySE7. Yes, there were other codes that pointed missfire on cylinder 1 & 6, but I didn't know what the PI300 was. Thanks for the reply.

ok. If you haven't changed plugs recently then do that and replace the wires too. 8mm Magnacore seems to be what the experts on this forum stand by.
If you've never changed plugs before start by hand tightening then to torque specs.
If you have done lots of plugs then apologies for stating the obvious

Actually, I have no clue how much torque I should use. Would you be so kind to enlighten me with that information please? I still don't know how/if I can get behind the engine to install the wires. It looks mighty tight. I'll be optimistic and keep telling myself others have done it, I should be able to as well.

Is therea trick to this? Certain easy parts that I can remove to assist with accessability?

It is a basic DIY, but your right its tight. Just take your time, maybe do half one day half the next. As for torque 20 Nm (15 lbf.ft)

As for plugs, for the 4.0 use Champion RC11 PYP B4 and for the4.6 use Champion RN11 YCC
I think Champion 322/RN11YC4 will also work. These are one option, just search plugs on this site as you will find many discussions about others (bosch etc.) opinions vary widely.

You need to be specific, what is the exact code, 1300 or a 1301???????????????????
1300 is a general emission misfire, 1301 is specific misfire code for cylinder #1.
