No, the key does not have to be on. Basically, if you have 12V at the large battery cable...you are replacing the need of the small wires job with the screwdriver jumping current across terminals.
If you have a test light, have someone turn the key to the crank position, while you check the small wire for 12V. If the small wire is carrying 12V as they turn the key, then ignition system is doing its job. And as long as you have a fully charged battery with clean, tight connections and good cables...you should have 12V at large cable on starter. With these two conditions met, starter should crank...if not...most likely starter has gone bad.
