Default How to perform a chemical O2 test?

Did a search and couldn't find anything in particular. It seems like a really silly question, but I'm stumped.

I purchased a tester which can either attach to a hand pump or a vacuum line on the truck. Not knowing which vacuum line to use( if any), I proceeded with using a hand pump. All I accomplished was sucking a bunch of coolant into the test vial. I followed instructions, which called for removing coolant from the tank before performing the test.

What am I doing wrong?

PS: I've attached a photo of what I'm working with. Thanks!
