Default Transmission Failure?

So I decided to upgrade my cooling system, had the radiator rodded, purchased a high flo 180 degree thermostat, got a used fan from Paul Grant, and put it all together yesterday. (I'm getting to the transmission part...). The good news is it seems my truck is running much cooler, but it's kind of hard to tell because my transmission is inop! After I got everything together last night I went to move it and it wouldn't go into gear, ah hah...I realized that I probably was low on fluid because of the internal cooler in the radiator, so I went and got some Dexron and put it in. It did move then, I drove it to the store but it wasn't right, down shifted too soon, slips. So I thought I might have overfilled it and this morning I drained some out to get the level perfect at the top in neutral with the transmission cool. Now it won't even budge. You guys that have owned these for years have any input? Why would removing and reinstalling the radiator have caused the transmission to fail? Seems totally disconnected.
